Abstract. . Locomotor activity rhythms were studied in eight species of nemobiine crickets with different habitat preferences. They showed similar patterns with a major peak of activity around dusk and a minor one around dawn. These patterns did not change within the natural range of daylengm (12–16 h). The acrophase time of the rhythm varied between -3.2 and +3.5 h as measured from dusk, and the free-running period (T) varied between 22.8 and 24.5 h. In the Dianemobius fascipes species complex these circadian parameters varied among nine local strains (8oS to 43oN) but did not show any latitudinal trend. In a temperate strain of this species complex (Dianemobius nigrofasciatus from Teshikaga, 43oN), two lines which differed from each other in mean T by 1.4h were selected, and a high heritability of T, 0.78 + 0.20 (mean±SE) was obtained. On the other hand, in a subtropical strain (Dianemobius fascipes from Ishigaki Island, 24oN), low heritabilities, 0.35 ± 0.15 for acrophase and 0.17 ± 0.21 for T, were obtained from the full-sib correlations. The variation in i seemed to be random and not related to fitness.  相似文献

Protein kinase C (PKC) is a multigene family of at least 12 isoforms involved in the transduction of extracellular signals. We investigated whether PKC-α, a major isoform known to be relatively abundant in brain tissue, is increased in human melanocytes relative to keratinocytes in vitro and in situ. Immunohistochemical staining for PKC-α in frozen neonatal human foreskin exhibited intermittent 2–3+ staining along the basal cell layer consistent with melanocytes, and 0–1+ staining of keratinocytes (on a scale of 0–3). Microscopic densitometry of the intermittent cellular staining was at least 3-fold greater than that of adjacent keratinocyte cell cytoplasm. Sequential frozen sections revealed similar intermittent cell staining with PKC-α and Mel-5 (tyrosinase related protein-1), known to specifically react with melanocytes. Northern blot analysis with a specific cDNA probe for PKC-α showed strong PKC-α mRNA expression in cultured melanocytes, whereas PKC-α mRNA in cultured non-stratifying keratinocytes was expressed at low levels. Western blot analysis revealed a prominent PKC-α band at approximately 80 kDa in melanocytes as opposed to a weak band in keratinocytes. Densitometry of the northern and western blots revealed that melanocytes had at least 10-fold more PKC-α mRNA and approximately 6-fold more PKC-α protein expression than keratinocytes. Total PKC activity measured in vitro revealed that melanocytes had 5-fold more activity than keratinocytes. The marked difference in melanocyte and keratinocyte expression of PKC-α provides further evidence for cell type specificity in the balance of PKC-α expression and may implicate differential PKC isoform signaling pathways in neuro-ectodermally derived cells.  相似文献

Melanoma among Japanese is rare, and differs in its clinical and histological characteristics from that found in Caucasians. In this study, the ganglioside expression of melanoma specimens obtained from Japanese was determined and compared with previously published data on Caucasians. The ganglioside composition of 25 biopsy melanoma specimens, including 13 primary and 12 metastatic lesions, was studied using thin layer chromatography. Four gangliosides (GM3, GD3, GM2, GD2) were most commonly expressed in melanomas in Japanese. The expression of gangliosides was quite variable in both primary and metastatic melanomas as seen in previous reports. No significant differences were observed between gangliosides from primary and metastatic sites. A new type of ganglioside expression, in which GM3 was nearly the only ganglioside (>95%), was found in metastatic tumors from two Japanese patients with acral lentiginous melanoma (ALM), which is the most common clinical and histopathological type of melanoma among Japanese but is very unusual among Caucasians. The patterns of expression were similar to those in Caucasians except for the detection of a “new” pattern.  相似文献
